Not too long. Glipizide ER has 100% bioavailability after a single oral dose. It will begin to release into your bloodstream 2 to 3 hours after taking it, and will reach maximum concentrations within 6 to 12 hours after dosing. The concentration levels will then remain constant throughout the 24 hour intervals in subsequent doses.

The typical starting dose for most patients is 5 mg daily, which should be taken orally with your first meal. Elderly patients or patients with liver disease may start on a lower dose of 2.5 mg. Based on how your body responds, your doctor may increase your dosage to a maximum daily dose of 20 mg.
